Overview

The tricuspid valve leaflets are thin flaps of fibrous tissue within the tricuspid valve, the heart structure separating the right atrium from the right ventricle. Classically, three leaflets are described—anterior, posterior, and septal—although anatomical studies show variability, with occasional additional or fewer leaflets in different individuals. Each leaflet is composed of connective tissue layers including the atrialis, spongiosa, fibrosa, and ventricularis, surrounded by endothelial cells. These leaflets tether via the chordae tendineae to papillary muscles in the right ventricle, functioning together to ensure one-way blood flow and prevent backflow during ventricular contraction. Pathology affecting the leaflets may lead to inadequate closure (regurgitation) or restriction (stenosis), contributing to cardiovascular disease. No specific drug directly targets the leaflets at the molecular level; interventions are structural or surgical.
