The TRIF-dependent signaling pathway refers to the series of molecular events initiated primarily by the engagement of Toll-like receptor 3 (TLR3) or Toll-like receptor 4 (TLR4), which recruit the adaptor protein TRIF (TIR domain-containing adaptor inducing IFN-β). TRIF stimulates downstream kinases and transcription factors, leading to the production of type I interferons (e.g., IFN-β), pro-inflammatory cytokines (such as TNF-α, IL-6), and the activation of nuclear factor kappa B (NF-κB) and interferon regulatory factors (IRFs). This pathway plays essential roles in the innate immune response, cell death (apoptosis and necroptosis), defense against viral infections, and inflammatory diseases. Since the pathway is not itself a protein or small molecule drug target, but rather a collection of molecular interactions, it does not directly correspond to a single therapeutic target
