Overview
TREM2 is a membrane glycoprotein and member of the immunoglobulin superfamily, primarily expressed on myeloid lineage cells. It forms a receptor complex with TYROBP/DAP12 or DAP10 adaptor proteins to mediate intracellular signaling. It regulates microglial survival and activation, phagocytosis, maintenance of brain homeostasis, and modulation of inflammatory responses. Variants are associated with Alzheimer's disease, Nasu-Hakola disease, and cancer, making it a therapeutic target.
