Triggering receptor expressed on myeloid cells-like protein 2 (TREML2) is a cell surface receptor belonging to the immunoglobulin superfamily, expressed predominantly on myeloid cells such as macrophages and microglia[4]. It is encoded by the TREML2 gene located on chromosome 6. TREML2 (also called TLT-2 or C6orf76) participates in the regulation of immune responses, including anti-inflammatory activities, and is involved in the modulation of myeloid cell function[4]. A missense variant in TREML2 is associated with protection against Alzheimer’s disease[4]. Unlike TREM2, current data do not support a role for TREML2 as a B7-H3 receptor[4]. TREML2 is studied for its role in neurodegenerative diseases and inflammation, but details on drug interactions, molecular inhibitors, or utility as a pharmacodynamic or predictive biomarker are not currently established in major databases[4]. While its homolog TREM2 is heavily investigated for Alzheimer’s disease and immuno-oncology, TREML2’s therapeutic relevance is less clearly defined, but it is considered a receptor with disease associations in the immune and neurodegenerative areas.
