Triglyceride synthesis in the liver is a central metabolic process by which hepatocytes convert fatty acids and glycerol into triglycerides (triacylglycerols, TG). This process is crucial for energy storage, lipid homeostasis, and the regulation of circulating lipids. The liver synthesizes triglycerides from both dietary sources and de novo synthesized fatty acids, then packages them into very-low-density lipoproteins (VLDL) for secretion into the bloodstream. Excessive hepatic TG accumulation underlies non-alcoholic fatty liver disease (NAFLD), while overproduction/secretion contributes to hypertriglyceridemia—a risk factor for cardiovascular disease. Selective hepatic insulin resistance results in continued TG production despite impaired suppression of gluconeogenesis—a hallmark feature observed in type 2 diabetes mellitus.
