TRIM28 interacting DNA damage repair enhancing non-coding transcript (TRIDENT) is a long non-coding RNA whose expression is upregulated by epidermal growth factor receptor (EGFR) activation, particularly in EGFR-driven non-small cell lung cancer. TRIDENT interacts with the protein TRIM28 and promotes its phosphorylation, which facilitates efficient repair of DNA damage in cancer cells. Knockdown of TRIDENT results in decreased tumor cell proliferation and increased sensitivity to multiple chemotherapeutic drugs. TRIDENT therefore represents a potential therapeutic target for overcoming drug resistance in certain cancers, as well as a possible biomarker for drug-resistant, EGFR-driven malignancies[5][3]. No drugs are currently known to directly target TRIDENT, but its inhibition sensitizes tumors to existing chemotherapies.
