Overview

Tripartite motif containing 40 (TRIM40) is a member of the TRIM protein family acting as an E3 ubiquitin-protein ligase with important roles as a negative regulator of inflammation and carcinogenesis, particularly in the gastrointestinal tract[1][2][5]. It limits the innate immune response by ubiquitinating and targeting receptors in the RLR (RIG-I-like receptor) pathway for degradation, thus dampening antiviral signaling[2][5]. It also promotes the neddylation of IKBKG/NEMO, contributing to stabilization of NFKBIA and inhibition of NF-kappa-B translocation and activity[1][5]. Research indicates TRIM40 interacts with ROCK1 and can inhibit cancer cell proliferation via the c-Myc/p21 axis, and its dysregulation may be implicated in gastrointestinal cancers and chronic inflammation[4]. No known therapeutics or biomarkers are currently associated with this target, and no main safety concerns are reported in the available literature[1][5].
