Overview

TRIM11 mRNA encodes the Tripartite motif-containing protein 11, an E3 ubiquitin-protein ligase that is a critical regulator of protein quality control and cellular homeostasis. In neurodegenerative diseases such as Alzheimer's and other tauopathies, TRIM11 acts as a neuroprotective factor by promoting the degradation of misfolded tau proteins and preventing their aggregation (Zhang et al., 2023, Science). Conversely, in various malignancies like glioma and breast cancer, TRIM11 mRNA is often overexpressed, where it functions as an oncogene by enhancing cell proliferation and suppressing apoptosis (Di et al., 2013; Chen et al., 2017). Therapeutic approaches targeting TRIM11 mRNA include the use of siRNA or antisense oligonucleotides to reduce its levels in cancer, or gene therapy and mRNA-based delivery to restore its function in the brain. Because of its dual role in promoting cell survival in cancer and clearing toxic proteins in the brain, therapeutic modulation requires careful consideration of tissue specificity and potential oncogenic risks (UniProt, 2024).
