Overview

Tripartite motif-containing protein 16 (TRIM16) is an atypical member of the TRIM family, characterized by two B-box domains (instead of the classical RING domain), enabling E3 ubiquitin ligase activity through RING-like folds[3][4][6]. TRIM16 serves as both a scaffold and a regulator, integrating protein ubiquitination with autophagic clearance of misfolded or aggregated proteins, especially under oxidative or proteotoxic stress[1][2]. It positively regulates the p62–KEAP1–NRF2 axis, stabilizing NRF2, and participates in cellular defense against proteinopathies, including neurodegenerative disease, cancer, and cardiac hypertrophy[1][2][5]. TRIM16 also interacts with other TRIM family members for diverse functions, including transcriptional regulation, cell differentiation, and immunity[3][5]. There are no currently known drugs that directly target TRIM16, but it is considered a potential therapeutic target due to its central role in proteostasis, cell stress responses, and disease-relevant proteinopathies[1][2][5].
