Overview

Tripartite motif-containing protein 28 (TRIM28), also known as KRAB-associated protein 1 (KAP1) or transcription intermediary factor 1-beta (TIF1-beta), is a multifunctional nuclear corepressor and chromatin regulator that interacts with the KRAB domain of zinc finger transcription factors to drive transcriptional silencing and heterochromatin formation[1][2]. TRIM28's structure includes a tripartite motif (RING finger, B-boxes, coiled-coil) characteristic of the TRIM family, as well as domains for interaction with heterochromatin protein 1 (HP1) and a bromodomain which recognizes acetylated lysine residues[2]. It plays critical roles in epigenetic silencing of endogenous retroviral elements, maintenance of genome integrity (notably in DNA double-strand break repair), and regulation of 3D chromatin architecture, notably in T cell activation and differentiation[3]. TRIM28 is essential for embryonic development and regulates genes involved in cell proliferation, survival, and immune responses such as suppression of viral gene expression. Aberrations in TRIM28 function or expression are linked to cancer progression, neurodegenerative disorders, viral infections, inflammation, and metabolism-related diseases[1][2][3][4]. As a consequence, it is an important, though challenging, candidate for therapeutic targeting in oncology, immunology, and virology.
