Overview

**Tripartite motif-containing protein 46 (TRIM46)** is a microtubule-associated protein of the TRIM family, characterized by zinc-binding domains—a RING finger, a B-box motif, and a coiled-coil region[3][7][8]. It plays a critical role in organizing parallel microtubule bundles in the proximal axon, which is essential for the specification of axons, regulation of neuronal polarity, and maintenance of efficient cargo delivery in neurons[1][2][3][5][8]. TRIM46 is highly enriched in the axon initial segment (AIS) and is central to early neural development, emerging before traditional axonal markers such as Tau1[2]. It is also associated with the proliferation of cancer cells, including those in lung and breast cancers[3]. However, there are currently no known direct drugs targeting TRIM46, nor is it established as a therapeutic target, receptor, enzyme, transporter, or transcription factor[3][8]. Loss of TRIM46 affects neuronal architecture and microtubule organization but does not lead to overt behavioral or structural deficits in knockout models, indicating functional redundancy or compensation in vivo[1][5].
