Tripartite motif-containing protein 66 (TRIM66) is a multifunctional E3 ubiquitin ligase and chromatin-associated epigenetic regulator belonging to the TRIM protein family. Structurally, it contains RING, B-box, coiled-coil, PHD, and bromodomain motifs, enabling it to recognize specific histone modifications (such as unmodified H3R2/K4 and acetylated H3K56ac)[1][3]. TRIM66 functions in transcriptional repression and regulates chromatin accessibility, playing critical roles in DNA damage repair by recruiting the SIRT6 deacetylase and thus ensuring genome integrity in embryonic stem cells and spermatids[1][3][4][5]. TRIM66 is low or undetectable in most tissues but is highly expressed in post-meiotic germ cells; aberrant overexpression is found in multiple cancers, where it promotes proliferation, metastasis, and poor prognosis, and serves as a potential diagnostic and prognostic biomarker[2][5]. No drugs currently target TRIM66 directly; however, its essential roles in DNA repair and chromatin regulation pose safety concerns for therapeutic inhibition[1][3].
