Overview

Tripartite motif-containing protein 66 (TRIM66) is a multifunctional E3 ubiquitin ligase and chromatin-associated epigenetic regulator belonging to the TRIM protein family. Structurally, it contains RING, B-box, coiled-coil, PHD, and bromodomain motifs, enabling it to recognize specific histone modifications (such as unmodified H3R2/K4 and acetylated H3K56ac)[1][3]. TRIM66 functions in transcriptional repression and regulates chromatin accessibility, playing critical roles in DNA damage repair by recruiting the SIRT6 deacetylase and thus ensuring genome integrity in embryonic stem cells and spermatids[1][3][4][5].
