Overview

Tripartite motif-containing protein 67 (TRIM67) is a Class I TRIM family E3 ubiquitin ligase that is predominantly expressed in the nervous system of mammals, where it regulates neuronal morphogenesis, cytoskeletal dynamics, exocytosis, and synapse maintenance[1][2][3]. TRIM67 interacts with several cytoskeletal and membrane trafficking proteins and is involved in processes critical for neuronal growth and neural circuit formation, such as axon branching, dendritic arborization, and filopodia stability[1][2]. Animal studies demonstrate that loss of TRIM67 leads to pronounced defects in brain structure, connectivity, and cognitive function, with TRIM67 knockout mice exhibiting hippocampal hypotrophy and impairments in spatial memory, social behavior, and sensorimotor gating[2]. TRIM67 does not have any current known drug modulators, nor is it an established therapeutic target in human disease, but it plays a significant role in brain development and function; disruptions may contribute to neurodevelopmental disorders[1][2][3].
