Overview

Tripartite motif-containing protein 72 (TRIM72, also called MG53) is a muscle-specific E3 ubiquitin ligase prominently involved in membrane repair by nucleating the assembly of the repair machinery at sites of plasma membrane injury[1][2][3][8]. It consists of a RING domain, B-box, coiled-coil, and C-terminal SPRY domain characteristic of the TRIM protein family[3]. TRIM72 is primarily expressed in striated muscle and the heart but is also found in smaller amounts in other tissues such as kidney, lung, and cornea[3]. Functional studies indicate that TRIM72 protects tissues during acute injury and ischemia, participates in innate immune signaling, influences metabolic regulation (including insulin resistance), and has been proposed to exert anti-tumor effects via its ligase activity[1][3]. Its role in rapidly promoting the repair of sarcolemmal and plasma membrane lesions makes it a candidate therapeutic target in muscle degenerative diseases and acute tissue injuries. Experimental therapies using recombinant MG53 are in development, but no approved drugs currently target TRIM72 directly[3].
