Overview

Tripartite motif-containing protein 74 (TRIM74) is a member of the tripartite motif (TRIM) protein family, defined by a conserved N-terminal RBCC motif consisting of a RING domain, one or two B-box domains, and a coiled-coil region. TRIM74 is a protein-coding gene located at 7q11.23. Like other TRIM proteins, it is predicted to act as an E3 ubiquitin ligase, contributing to ubiquitin-mediated protein modification and degradation. The functional significance of TRIM74 in human biology or disease remains largely uncharacterized in the literature; no direct evidence links it to specific pathologies or defines it as a recognized therapeutic target. TRIM74 also appears under the name TRIM50C, although this can cause confusion with related TRIM genes and highlights nomenclature overlap within the family. TRIM74 is not established as a therapeutic target (such as an enzyme, receptor, transporter, or channel) in clinical drug discovery. Its precise molecular function, interacting partners, and roles in disease await further elucidation. Occasional references describe TRIM74 mutations potentially impacting protein homeostasis in models of neurodegeneration, suggesting a role in proteostasis, but these findings are preliminary and not confirmed in human disease. Major drug and biomarker databases do not list drugs or biomarkers directly related to TRIM74.
