tRNA (guanine(37)-N(1))-methyltransferase (TRMT5) is an enzyme responsible for methylating the N1 position of guanosine-37 (G37) in various tRNAs, a critical post-transcriptional modification necessary for stable and functional tRNA structure, especially in mitochondria[3][4]. Deficiency or mutation in TRMT5 disrupts mitochondrial tRNA modification, leading to defective mitochondrial protein synthesis and is associated with multiple mitochondrial respiratory-chain deficiencies[2][3][4]. There are currently no drugs specifically known to target TRMT5 in clinical practice.
