Overview

tRNA methyltransferase 10 homolog B (TRMT10B) is an enzyme belonging to the TRM10 methyltransferase family, conserved in Eukarya and Archaea, that specifically catalyzes methylation of adenosine at position 9 (m^1A9) of a single nuclear-encoded tRNA, notably tRNA^Asp-GTC^, in human cells[1][2][3]. Unlike its paralog TRMT10A, which methylates guanosine at the same position on multiple tRNA species, TRMT10B demonstrates remarkable substrate specificity, acting only on tRNA^Asp-GTC^ and not compensated by TRMT10A[1][2][3]. This methylation is believed to contribute to the structural stability and appropriate functioning of tRNA, thus indirectly influencing protein synthesis. TRMT10B is localized in the cytosol and is associated with the TIM22 mitochondrial import inner membrane insertion complex[1][7]. There are currently no known drugs that specifically target TRMT10B, and no established role as a disease biomarker or therapeutic target. Disruption of related tRNA methyltransferases, but not specifically TRMT10B, has been linked to certain human diseases, suggesting its modification activities may be important for cellular homeostasis[2].
