tRNA methyltransferase 13 (TRMT13) is a bifunctional enzyme involved in post-transcriptional modification of tRNA and gene regulation. In the cytoplasm, TRMT13 catalyzes the 2'-O-methylation of specific tRNAs, thereby fine-tuning protein synthesis by modulating translation[1][2]. Independently of its methyltransferase activity, nuclear TRMT13 binds directly to DNA at promoters via its CHHC zinc finger domain, acting as a transcriptional co-activator for genes controlling cell migration, particularly factors involved in epithelial–mesenchymal transition (EMT)[1][2]. Overexpression of TRMT13 correlates with poor prognosis in several cancers, highlighting its dual epitranscriptomic and transcriptional regulatory roles as well as its significance as a prognostic marker and a potential therapeutic target[1][2].
