Overview

tRNA wybutosine-synthesizing protein 3 homolog (TYW3) is a protein-coding gene found across eukaryotes and archaea but not bacteria. It encodes a probable S-adenosyl-L-methionine-dependent methyltransferase that participates in the biosynthetic pathway of wybutosine (yW), a highly modified guanosine with a tricyclic base located at position 37 adjacent to the anticodon of eukaryotic phenylalanine tRNA. TYW3 catalyzes a key methylation step, using S-adenosyl-L-methionine as a methyl donor, and is essential for the maintenance of translational reading-frame fidelity on the ribosome. Mutations in TYW3 are rarely implicated in human disorders; no evidence supports its use as a therapeutic target, biomarker, or drug-binding protein.
