Overview

Trophinin is a transmembrane cell adhesion protein encoded by the TRO gene, originally characterized by its role in mediating apical cell adhesion between trophoblastic cells and the luminal epithelial cells of the endometrium, enabling the initial attachment of the embryo during implantation. It is a member of the MAGE (Melanoma-associated antigen) superfamily, specifically the MAGE-D subfamily, and displays structural similarity to other proteins of this family. Alternative splicing yields multiple transcript variants, including the full-length KIAA1114 and shorter forms. In addition to its physiological function in embryo implantation, trophinin/KIAA1114 has been implicated in the identification and propagation of tumor-initiating cells in hepatocellular carcinoma and potentially other cancers, serving as a candidate biomarker whose expression correlates with tumorigenic capacity and TIC hallmarks; however, it is not broadly targeted by approved drugs and its clinical targeting is experimental at present. Its expression is generally restricted in adult tissues, but notable in testis, kidney, and brain.
