Trophoblast cell-surface antigen 2 receptor (TROP2) is a membrane-bound glycoprotein encoded by the TACSTD2 gene on chromosome 1p32. It functions as a cell surface receptor involved in self-renewal, proliferation, and transformation of cells. In embryonic development it plays critical roles in placenta formation and organogenesis. In adults it is overexpressed in many epithelial-derived tumors such as breast, urothelial, prostate, pancreatic, ovarian cancers as well as certain lymphomas. Overexpression or deregulation of TROP2 correlates with aggressive disease features like advanced stage or nodal metastasis depending on tumor type. It is the validated therapeutic target for sacituzumab govitecan—an antibody-drug conjugate approved for triple-negative breast cancer and urothelial carcinoma—and serves both as a therapeutic target and prognostic biomarker across multiple malignancies
