Overview

Trophoblast cell-surface marker 2 (TROP2), also known as tumor-associated calcium signal transducer 2 (TACSTD2), is a type I transmembrane glycoprotein that functions as a cell-surface receptor and signal transducer. It plays a critical role in regulating cell proliferation, adhesion, and migration through various signaling pathways, including the MAPK/ERK and calcium-mediated pathways. TROP2 is highly overexpressed in a wide range of epithelial carcinomas, including breast, lung, urothelial, and colorectal cancers, while showing limited expression in normal tissues. This differential expression makes it an ideal therapeutic target, particularly for antibody-drug conjugates (ADCs). The most prominent drug targeting this molecule is sacituzumab govitecan, which has been approved for triple-negative breast cancer and urothelial cancer. Ongoing research continues to explore TROP2 as a biomarker for prognosis and a target for novel therapies in multiple solid tumor types.
