Overview

Trophozoite export protein 1 (Tex1) is a large, highly conserved protein expressed during the asexual blood stage of the malaria parasite Plasmodium falciparum. It is exported from the parasite to the Maurer's clefts, which are parasite-derived organelles in the host erythrocyte cytoplasm involved in protein trafficking and host cell remodeling. Tex1 contains two major antigenic regions: a coiled-coil domain known as P27 and an intrinsically unstructured region known as P27A. These regions are recognized by antibodies from individuals living in malaria-endemic areas, and these antibodies are associated with clinical protection. The P27A fragment has been developed as a synthetic peptide vaccine candidate and has undergone Phase 1 clinical testing, demonstrating safety and the ability to induce antibodies that inhibit parasite growth through antibody-dependent cellular inhibition (ADCI).
