Overview

Tropomodulin-3 is a member of the tropomodulin family of actin-regulatory proteins encoded by the TMOD3 gene. It regulates actin filament dynamics and organization by capping the pointed (minus) ends of actin filaments, preventing both polymerization and depolymerization. This capping activity is essential for defining actin filament length, cell shape, motility, adhesion, and maintenance of cytoskeletal architecture. TMOD3 is broadly expressed in non-muscle cells, as well as in specialized cell types—such as endothelial, epithelial, neuronal, and erythroid cells—where it stabilizes actin–tropomyosin filaments and contributes to processes including lamellipodia protrusion, cell migration, and assembly of adherens junctions. TMOD3’s function is critical for development, cellular homeostasis, and certain disease pathologies. Altered expression or activity has been implicated in cancer progression, chemoresistance, impaired erythropoiesis, and neurodevelopmental disorders[1][2][3][4][5].
