Tropomyosin-related kinase B (TRKB) is a receptor tyrosine kinase that plays a crucial role in neuronal survival, differentiation, and synaptic plasticity. It is a high-affinity receptor for BDNF, NT4/5, and NT3. Activation of TRKB initiates multiple downstream signaling cascades including Ras-MAPK/ERK, PI3K-Akt, and PLCγ. Aberrant activation or overexpression of TRKB/NTRK2 has been implicated in various diseases, including cancer and psychiatric conditions.
Selective TRKB inhibitors and Pan-Trk inhibitors block the kinase activity of TRKB, inhibiting downstream signaling pathways such as Ras-MAPK/ERK, PI3K-Akt, and PLCγ.
