Overview

TRPM8 channel-associated factor 1 (TCAF1), also known as FAM115A, is an intracellular regulatory protein that binds directly to the TRPM8 ion channel, a cold sensor and regulator of cellular processes such as migration. TCAF1 promotes the trafficking of TRPM8 to the plasma membrane and modulates the channel’s gating, typically antagonizing cell migration—especially in prostate cancer models. Its activity contrasts with the related protein TCAF2, and loss of TCAF1 expression is associated with increased metastatic behavior in prostate cancer. In addition to ion channel regulation, TCAF1 has roles in protecting cells under replication stress by regulating TRPV2-mediated calcium release, which is critical for replication fork stability and genomic integrity. TCAF1 has undergone evolutionary adaptation in humans, including structural diversification and copy number variation, suggesting broader physiological significance[1][2][3]. No direct drugs target TCAF1, but its interaction with TRPM8 suggests its abundance or activity could alter the response to TRPM8 modulators (such as menthol or icilin). TCAF1’s expression profile and regulatory function make it a potentially useful biomarker in prostate cancer prognosis and replication stress responses. Summary of key molecular and biological points: - Intracellular regulatory/adaptor protein that modulates ion channel trafficking and function. - Binds most notably to TRPM8 and TRPV2 channels, influencing their localization and activity. - Functions in cell migration (especially anti-migratory in cancer contexts) and replication stress response. - May serve as a biomarker for prostate cancer progression and replication stress.

Mechanism of action

No established drug mechanism of action for TCAF1 itself. TCAF1 can alter the membrane localization and gating properties of TRPM8—so, compounds modulating TRPM8 might have altered effects depending on TCAF1 expression or activity[2][3].
