Truncated Trophoblast cell-surface antigen 2 (tTrop2) is a cancer-specific neo-epitope formed by the proteolytic cleavage of the full-length Trop-2 protein, often at the R87-T88 site. While full-length Trop-2 is expressed in various normal epithelial tissues, the truncated or cleaved form is predominantly found on the surface of tumor cells, where it promotes oncogenic signaling and cell proliferation. This tumor-specific display makes tTrop2 an attractive target for precision therapies, including antibody-drug conjugates (ADCs) and chimeric antigen receptor (CAR) T-cell therapies, as it minimizes off-target effects on healthy tissues. CAR-T cells directed against the truncated extracellular domain of Trop-2 are designed to recognize and eliminate cells expressing this specific molecular signature. Research into tTrop2-targeted modalities, such as the antibody Hu2G10 and the ADC LCB82, highlights its potential to overcome resistance pathways associated with conventional Trop-2-targeted agents.
