Trypanothione reductase is an essential flavoenzyme unique to kinetoplastid protozoa that maintains cellular redox balance via reduction of trypanothione disulfide using electrons from NADPH through its FAD cofactor. Its absence or inhibition leads to parasite death due to oxidative damage. Because there are no close human homologs, it represents an ideal selective drug target against diseases like leishmaniasis and Chagas' disease. Multiple structural studies have revealed both active site and novel allosteric pockets suitable for rational drug design efforts.
