Overview

Tryptase beta-2 is a serine protease enzyme predominantly expressed and secreted by mast cells, functioning as a major neutral protease released during mast cell degranulation[1][2][3][8]. It is typically only active as a heparin-stabilized tetramer and is resistant to known endogenous proteinase inhibitors[1][2]. Tryptase beta-2, encoded by the TPSB2 gene, is involved in mediating allergic and inflammatory responses and is implicated in the pathogenesis of asthma and other inflammatory diseases. Its enzymatic activity also plays a role in activating influenza A virus within the upper respiratory tract[1]. High tryptase levels can serve as a biomarker for mast cell activation in clinical diagnostics. Genetic and functional diversity exists between beta-tryptase isoforms, and the enzyme is a potential therapeutic target for conditions involving mast cell dysfunction.
