Overview

Tryptase delta 1 is a trypsin-like serine protease encoded by the TPSD1 gene on human chromosome 16p13.3. It belongs to the neutral mast cell tryptase family (peptidase family S1) and is released during mast cell degranulation, often in response to allergic or inflammatory stimuli. Structurally, it typically forms tetramers stabilized by heparin, remaining active only in this conformation. Its functions include regulating cell proliferation and growth, processing (pro)hormones, activating fibrinolytic enzymes, and modulating inflammatory and immune responses. Tryptase delta 1—along with other tryptases—is implicated in the pathogenesis of allergic diseases such as asthma, and may play roles in hereditary conditions linked to altered mast cell activity. Drugs targeting tryptase delta 1 focus on inhibiting its enzymatic actions to reduce inflammation, but face challenges due to the enzyme's importance in normal physiology and genetic diversity among patients
