Overview

Tryptase gamma 1 (TPSG1) is a membrane-associated serine protease encoded by the human TPSG1 gene, located on chromosome 16p13.3[1][3][4]. It is a member of the tryptase family (peptidase family S1), with a distinctive C-terminal hydrophobic domain for membrane anchoring, unlike other secretory tryptases[1][3]. TPSG1 is mainly associated with mast cells and, to a lesser extent, basophils, and is implicated in proteolytic cleavage of substrates during immune and allergic responses[1][3][4]. Tryptase gamma 1 is active as a heparin-stabilized tetramer and is resilient to most endogenous inhibitors[3][4]. It has been linked to the pathogenesis of asthma, allergic inflammation, and related disorders[1][4][6]. Recent evidence suggests a possible role in tumor cell migration and invasion, such as in breast cancer[6].
