The TPSP1 gene is classified as a pseudogene, meaning that it is a non-functional segment of DNA that shares homology with protein-coding tryptase genes but does not produce a functional protein product[1]. Unlike its protein-coding relatives—such as tryptase alpha, beta, delta, and gamma—TPSP1 does not play a direct biological or clinical role, nor is it considered a receptor, enzyme, or drug target[1]. In contrast, functional tryptase genes (TPSAB1, TPSB2, TPSD1, TPSG1) encode serine proteases involved in mast cell biology and are sometimes clinically relevant as biomarkers or drug targets in conditions like anaphylaxis and mastocytosis[2][3]. The existence of tryptase pseudogenes like TPSP1 reflects gene family evolution, but these pseudogenes themselves do not have a direct functional or pathological role[1].
