Overview

Tryptophan 5-hydroxylase 2 (TPH2) is a neuron-specific isoform of tryptophan hydroxylase, the rate-limiting enzyme in serotonin biosynthesis. It catalyzes the conversion of L-tryptophan to 5-hydroxytryptophan (5-HTP) in the brain. Serotonin synthesized via this pathway plays critical roles in mood regulation, stress response, aggression control, and sleep-wake cycles. Genetic variations affecting its activity are associated with psychiatric disorders.
