Overview

TSBP1 and BTNL2 antisense RNA 1 (TSBP1-AS1) is a human long non-coding RNA (lncRNA) gene located at chromosome 6p21.32 within the major histocompatibility complex (MHC) region. It is classified as an antisense RNA because its transcript is complementary to the TSBP1 and BTNL2 loci, and it is believed to play a role in the cis- or trans-regulation of these and potentially other nearby genes. TSBP1-AS1 is not a protein or traditional drug target, but rather a regulatory RNA molecule acting primarily at the level of transcription or epigenetic modification, influencing the expression of genes in its vicinity. Disease-association studies have linked genomic variants within the TSBP1-AS1 locus to a wide range of complex disorders, notably immune-mediated diseases (such as sarcoidosis and Graves disease), metabolic disorders (type 2 diabetes, hypertension), and infectious disease outcomes (HIV-1 progression and resistance). There are no drugs directly targeting TSBP1-AS1 or its transcript; its significance is mainly as a locus of genetic variation impacting disease risk and potentially modifying gene regulation in the MHC region.
