TSR2 ribosome maturation factor is a highly conserved protein involved in eukaryotic ribosome biogenesis, specifically acting as a chaperone—also termed an escortin—for the ribosomal protein eS26 (Rps26)[1][2][3]. After ribosomal proteins are imported into the nucleus, TSR2 mediates the dissociation of eS26 from its nuclear importins by a RanGTP-independent mechanism, shields it from aggregation and proteolysis, and traffics it to the pre-ribosomal 90S complex during the early stages of ribosome assembly[1][2]. TSR2 can also extract Rps26 from mature ribosomes under certain stress conditions, temporarily storing it to allow ribosome remodeling and repair, thereby facilitating cellular adaptation to stress[2]. In yeast and humans, depletion or mutation of TSR2 leads to defects in ribosome maturation, reduced cell growth, and is linked functionally (but not genetically) to Diamond-Blackfan anemia through its client protein eS26[1][2][3]. TSR2 does not belong to any classical druggable target class (receptor, enzyme, transporter), and there are as yet no recognized drugs or therapeutic programs targeting this factor.
