Overview

TtgR is a key transcriptional repressor in Pseudomonas putida that belongs to the TetR family of regulators. It plays a critical role in bacterial survival by controlling the expression of the TtgABC multidrug efflux pump, which expels a diverse array of toxic substances including antibiotics, organic solvents, and plant secondary metabolites (UniProt P0C0V2). TtgR binds as a homodimer to the operator DNA, sterically hindering RNA polymerase and preventing transcription of the efflux system (Alguel et al., 2007). Upon binding to various ligands such as chloramphenicol or flavonoids, TtgR undergoes a conformational change that triggers its dissociation from the DNA, thereby inducing the expression of the efflux system (Teran et al., 2003). This regulatory mechanism allows the bacteria to rapidly adapt to the presence of environmental toxins and antimicrobial agents. Because it regulates a broad-spectrum resistance mechanism, TtgR is a significant target for research into antimicrobial resistance and the development of efflux pump inhibitors to restore antibiotic efficacy. Inhibiting TtgR or preventing its dissociation could potentially sensitize resistant strains to existing antimicrobial treatments.
