Overview

TTK protein kinase, also known as Monopolar spindle 1 (MPS1), is a dual-specificity kinase that plays a critical role in the spindle assembly checkpoint (SAC), ensuring proper chromosome segregation during mitosis. It is highly expressed in rapidly proliferating cells and is significantly overexpressed in a wide range of human malignancies, including breast, lung, and pancreatic cancers, while showing limited expression in normal adult tissues except for the testis. This tumor-associated expression pattern makes TTK-derived epitopes attractive targets for cancer immunotherapy, particularly peptide-based vaccines. In a therapeutic context, specific TTK-derived peptides (epitopes) are used to prime the immune system to recognize and destroy cancer cells. When these epitopes are presented by Major Histocompatibility Complex (MHC) molecules on the surface of tumor cells, they can be recognized by cytotoxic T lymphocytes (CTLs). Clinical strategies involving TTK-derived epitopes often focus on HLA-restricted peptide vaccines designed to elicit a robust, tumor-specific immune response while minimizing damage to healthy somatic tissues.
