Tubby-like protein 4 (TULP4) is a member of the tubby superfamily present in vertebrates and is structurally characterized by a conserved C-terminal tubby domain and a variable N-terminal region that—unique among tubby family members—contains WD40 repeats and a SOCS (suppressor of cytokine signaling) box domain[1]. This structural organization allows TULP4 to function as part of a novel E3 ubiquitin ligase complex, interacting with CUL5, elongin B/C, and RNF7, implicating TULP4 in protein ubiquitination[1][3]. TULP4 is localized to the cytoplasm[1], and studies in mice suggest a role in neurodevelopment by influencing neuronal migration; rare TULP4 gene variants have been associated with schizophrenia-related phenotypes[3]. Its exact cellular substrates and pathophysiological mechanisms remain unclear. There is currently no information indicating TULP4 is a direct therapeutic target or that it interacts with any approved drugs; it lacks established biomarker or safety concern status[3][6].
