Tubby-related protein 3 (TULP3) is a specialized adapter protein that plays a fundamental role in the trafficking of membrane-associated proteins, particularly G protein-coupled receptors (GPCRs), into the primary cilium [1]. It functions by linking the intraflagellar transport complex A (IFT-A) to phosphoinositides, specifically PI(4,5)P2, on the ciliary membrane, thereby facilitating the entry of signaling molecules like GPR161 and Somatostatin receptor 3 [2]. TULP3 is a critical regulator of the Hedgehog (Hh) signaling pathway, where it typically acts as a negative regulator by ensuring the ciliary localization of Hh inhibitors [3]. Mutations or dysregulation of TULP3 are linked to various ciliopathies, including polycystic kidney disease and developmental malformations, as well as certain cancers where it may promote epithelial-mesenchymal transition and tumor stemness [4]. While no small molecule drugs are currently approved for TULP3, its mRNA is a target of interest for RNA-based therapies such as siRNA and antisense oligonucleotides designed to modulate its expression in disease contexts [5].
