Overview

Tubulin beta-2B chain (TUBB2B) is a critical structural component of microtubules, which are dynamic polymers essential for cell division, intracellular transport, and the maintenance of cell architecture (UniProt Q9BVA1). TUBB2B is particularly prominent in the developing brain, where it facilitates the migration of neurons and the proper formation of the cerebral cortex (NCBI Gene ID: 347733). Pathogenic variants in the TUBB2B gene lead to a spectrum of neurodevelopmental conditions known as tubulinopathies, characterized by brain malformations such as polymicrogyria, pachygyria, and microcephaly (OMIM 612850). In the context of oncology, beta-tubulins are the molecular targets for several classes of antimitotic drugs, including taxanes (e.g., paclitaxel) and vinca alkaloids (e.g., vincristine), which disrupt microtubule dynamics to induce cell cycle arrest (PubMed PMID: 25130172). While current therapies primarily target the protein product, the TUBB2B mRNA is an emerging focus for precision medicine, with research exploring antisense oligonucleotides and other RNA-targeted strategies to address the underlying genetic causes of tubulin-related cortical malformations.
