Tubulin beta chain (TUBB)

Target
TUBB
Molecular classification
Cytoskeletal protein, Microtubule subunit, GTPase (Tubulin/FtsZ family)[4][5]
01

Overview

The tubulin beta chain is a major structural protein that forms heterodimers with alpha-tubulin to create the building blocks of microtubules. Microtubules are dynamic cytoskeletal filaments essential for maintaining cell shape, enabling intracellular transport, segregating chromosomes during mitosis/meiosis, and facilitating cellular motility. The protein binds GTP at specific sites critical for its polymerization dynamics. Because of its central role in cell division and structure, the tubulin beta chain is a validated therapeutic target—especially in oncology—where drugs that disrupt its function are widely used as chemotherapeutics.[1][3][5][6]

Mechanism of action

Drugs targeting tubulin beta chain generally act by binding to tubulin and disrupting microtubule dynamics, either stabilizing or destabilizing microtubules. This interferes with mitosis and leads to cell cycle arrest and apoptosis in rapidly dividing cells such as cancer cells.[4]

Safety considerations

Therapeutic targeting of tubulin can cause side effects such as neurotoxicity, myelosuppression, peripheral neuropathy, gastrointestinal disturbances, and alopecia due to the essential role of microtubules in normal cellular functions.
