Tubulin delta-1 (TUBD1) is a noncanonical member of the tubulin superfamily that functions as a structural component of the cytoskeleton, particularly involved in the assembly and stability of centrioles and cilia[1][2][3][4]. It participates in forming compound microtubule structures within centrioles, is essential for proper microtubule organization, and acts as a positive regulator of hedgehog signaling[1][2]. TUBD1 is required for centriole architecture, stable recruitment of centriole-associated scaffolding proteins, and correct cell cycle progression of centrioles[1]. In mammalian spermatogenesis, TUBD1 stabilizes kinetochores, is essential for spindle polarity and cytokinesis, and works with microtubule-severing enzymes to shape structures essential for sperm head formation, with knockout models showing resultant infertility[3]. It is associated with diseases involving ciliary dysfunction and some congenital heart defects[2][3]. There are currently no known drugs that directly target TUBD1, and it is not presently considered a therapeutic target.
