Overview

Tubulin epsilon and delta complex protein 1 (TEDC1) is a protein localized to centrioles, functioning as a critical positive regulator of ciliary hedgehog signaling and essential for centriole stability[1][2][6]. TEDC1 physically interacts with TEDC2, delta-tubulin (TUBD1), and epsilon-tubulin (TUBE1), forming a tetrameric complex fundamental for centriole structure and assembly[2][3][6]. Loss of TEDC1 phenocopies the loss of delta-tubulin and epsilon-tubulin (aberrant centrioles, lacking triplet microtubules, and prone to disintegration), reflecting its direct role in architectural maintenance—not signaling or catalysis[2][4]. While associated with some congenital diseases such as lacrimal duct obstruction and microcephaly, TEDC1 is not exploited as a drug target nor is it a recognized biomarker or therapeutic safety concern[1]. Its primary biological role is in the foundational assembly and function of centrioles and cilia in eukaryotic cells[2][5].
