Overview

TEDC2 is a nuclear protein containing a domain of unknown function (DUF4693), localizing mainly in centrioles and cilia. It forms a structural complex with TEDC1, delta-tubulin (TUBD1), and epsilon-tubulin (TUBE1), all of which are necessary for centriole integrity and assembly; loss of TEDC2 disables triplet microtubule formation and undermines mitotic progression. TEDC2 is widely expressed in normal and cancerous tissues, with upregulation correlated to poor prognosis and reduced immune cell infiltration in lung adenocarcinoma and other cancers. Knockdown of TEDC2 impedes tumor cell proliferation and migration, suggesting its regulatory role in the cell cycle, and highlighting it as a potential biomarker, though not a curated drug target as of now. TEDC2 interacts with proteins involved in DNA replication and centriole function (e.g., CDC45, TUBE1, FEZ1), and may be transcriptionally regulated by tumor suppressor elements (WT1, MZF1, P53).
