Tubulin epsilon chain (TUBE1) is a *non-canonical* member of the tubulin superfamily, distinct from alpha-, beta-, and gamma-tubulins. It localizes to the centrosome and basal bodies, where it is fundamental for the assembly and stability of triplet and doublet microtubules, especially in centrioles and ciliary structures[1][2][3][4]. TUBE1 forms complexes with delta-tubulin (TUBD1) and other proteins (TEDC1, TEDC2), which are collectively required for robust centriole architecture and microtubule integrity[2]. Loss of TUBE1 in mammals results in defective meiotic spindle formation, impaired sperm tail (axoneme) construction, abnormal sperm head morphology, and male sterility[1]. In lower eukaryotes, absence of epsilon-tubulin disrupts formation of flagella/cilia and leads to centriole disintegration[1][3]. Structural studies suggest it may stabilize microtubules as a capping protein or regulate microtubule junction formation[3].
