Tubulin gamma-1 chain (TUBG1) is a highly conserved component of the tubulin superfamily essential for microtubule nucleation. TUBG1, together with associated proteins (such as GCPs—gamma-complex proteins), forms the gamma-tubulin ring complex (γ-TuRC), a multi-protein assembly that serves as a template for the initiation (nucleation) of new microtubules at microtubule organizing centers (MTOCs), particularly the centrosome[1]. This process is critical for accurate mitotic spindle formation, cell division, and many fundamental cellular processes such as organelle positioning and intracellular transport[1][2]. γ-Tubulin has additional roles in regulating microtubule dynamics, controlling E2F-driven transcription in the nucleus, and participating in DNA damage responses[1][3]. Defects in TUBG1 function or regulation have been implicated in certain cancers and neurodevelopmental conditions, but the protein is not currently considered a direct pharmacological or therapeutic target due to its essential, ubiquitous role in cell biology and the lack of selective modulators[1][3][4].
Not applicable for drugs, as there are no known direct pharmacological agents targeting TUBG1. (Experimental approaches may include disruption by genetic or protein–protein interaction inhibitors, but these are not clinical drugs.)[1][3].
