TTLL10 encodes an enzyme responsible for elongating polyglycine chains on already monoglycylated substrates. Unlike TTLL8, which initiates monoglycylation, TTLL10 only adds additional glycine residues to those branches, in both α- and β-tubulin tails, and also modifies NAP1, particularly in spermatids. TTLL10 activity requires pre-existing monoglycylation, is enhanced by tubulin glutamylation, and self-limits its association with microtubules as chain length increases. TTLL10 is expressed predominantly in testis (mouse), with possible roles in gametogenesis through NAP1 modification. TTLL10 is not an established drug target and has no known interacting drugs. Its activity contributes to post-translational modification patterns required for microtubule and chromatin function. Key distinctions: TTLL10 should not be confused with TTLL5, which is a separate enzyme in the same family. TTLL10 is not a therapeutic target nor is it associated with clinical biomarker or pharmacological safety data.
