Tubulin tyrosine ligase-like protein 2 (TTLL2) is an enzyme belonging to the tubulin tyrosine ligase-like family, predicted to catalyze the generation of polyglutamate side chains on the gamma-carboxyl group of specific glutamate residues in the C-terminal tail of target proteins, notably tubulins[1][7]. TTLL2 likely participates in the post-translational polyglutamylation of tubulin, a modification crucial for microtubule function and organization[1][7]. It may require interaction with other protein partners for full enzymatic activity and is implicated in the initiation of polyglutamylation rather than its elongation[7]. TTLL2 is associated with some cancers, including large intestine adenocarcinoma, but specific therapeutics targeting TTLL2 have not been reported[1]. Its expression and functional relevance have also been noted in testis and ciliary basal body, suggesting roles in cilia function and spermatogenesis[1][4].
