Overview

Tudor domain-containing protein 5 (TDRD5) is an RNA-binding protein that plays an essential role in germline development, retrotransposon repression, and piRNA-mediated gene silencing during spermatogenesis. It is required for proper assembly of chromatoid bodies and pi-bodies in male germ cells, and is crucial for the production and processing of pachytene piRNAs, which help prevent mobilization of transposable elements and protect genome integrity in germ cells[1][2][5]. Loss of TDRD5 disrupts piRNA biogenesis, transposon silencing, chromatoid body structure, and leads to spermatogenic arrest and infertility in males[1][2][5]. TDRD5 is expressed in germ cells of both testes and ovaries, though its most defined function is in spermatogenesis and male fertility[2][5]. No clinically approved drugs target TDRD5, and it is not currently considered a therapeutic target for somatic diseases.
