The **tumor antigen-HLA complex** refers to the molecular assembly in which a tumor-derived peptide (antigen) is bound and presented by a human leukocyte antigen (HLA) molecule on the surface of a cell, facilitating recognition by T cell receptors (TCRs). This complex is central to adaptive immunity and underpins many modern cancer immunotherapies, including TCR-based therapies and peptide vaccines. Tumor antigen-HLA complexes mediate immune targeting of tumor cells but can also be involved in immune evasion when tumors disrupt antigen presentation machinery or lose HLA expression. Optimal immunotherapies require precise identification of both the presenting HLA allele and the tumor peptide, as the immune response is highly specific to this complex.
